Vector construction tasks can be requested and fulfilled in the Protein Production module.
Vector construction tasks can be registered manually on the Vector Construction page or automatically, when a Protein Request has been made but the system does not have vectors registered to express that protein.
Manual vector construction requests can be registered by DNA, AA, or VRP/Isotype. When requesting by DNA or AA sequence, the user will be presented with a form for registration. Each sequence request can optionally be associated with a project. Requesting by variable region pair and isotype will allow the user to optionally select a project, then select any number of VRP’s and any number of isotypes. A vector construction request will be registered for each unique combination of VRP/Isotype.
Any email addresses specified in the Application Settings' Vector Construction Request Email List will be notified whenever a new request is created.
Vector Construction Task Fulfillment
To fulfill one or more vector construction requests, select their checkboxes on the left and then click Fulfill. A wizard will appear which has two steps. The first displays the request ID, the requested sequence, and the requestor, as well as a Plasmid column. The plasmid column contains any plasmids already registered which can satisfy the given request. If there are no plasmids registered, the user will need to register new plasmids to fulfill the vector construction request - click Next to do so.
The second step of the wizard is only necessary if there are one or more requested vectors which were not able to be fulfilled in step one. The form in step two has the same columns as standard plasmid registration, with the addition of the optional DNA Recovered and Size fields. If these are specified, lots of the newly registered plasmid are automatically registered after submission. The only required fields for fulfillment of a new plasmid are a name and the Coding DNA Sequence, which must translate into the requested vector.
Clicking Fulfill closes the wizard and completes the vector construction tasks as necessary. If the task was automatically generated by a protein request task, that task will now become unblocked, and will appear in the Express page.
