Antibodies can be registered by sequence, plasmid, clone, variable region, or variable region pair in the materials module

Antibody Registration

To register antibodies in Affinity, click on Register Antibodies in the upper left of the Antibody index page, then select from sequences, plasmids, clones, variable regions, or variable region pairs.

When registering from clones or variable regions, rather than entering the data in the standard spreadsheet input, users select one or more clones (or one or more variable regions) and then a heavy and light chain isotype. This will then create an antibody for each combination (i.e. if 3 clones are selected, 3 antibodies with the selected HC and LC Isotypes will be registered).

Multispecifics are a type of antibody which are not limited to the traditional single HC/LC combination. To register a multispecific antibody, click Register Multispecifics and then select either From plasmids or From sequences. This selection affects which information is required for registration, either a series of sequences or a series of plasmid selections.

Multispecific antibodies require two heavy chains, have the option for up to four light chains during registration, and also have fields for specification of valency and multispecific type.

Antibody Index and Details

The index page for antibodies allow users to review the existing antibodies registered in the system. Filters can be applied on any column to narrow the displayed antibodies. Click on an antibodies' ID to navigate to its details page.

The details section allows users with proper permissions to edit the name, description, phase, format, valence, and multispecific type of an antibody. It will also display the calculated attributes, such as Molecular Weight, Extinction Coefficient, and Isoelectric Point.

In the top right of the page, the user is able to tag, comment, and attach any files to the entity.

Lots of the selected antibody are listed on the details page and the user can register new lots by clicking the Register Lots button.

Select an assay type in the Assay Results section of the details page to display all assays of that type against samples of the selected antibody.

Beneath the Assay Results, the user can view and edit the Targets associated with this antibody. Clicking Add target to select either a target or a material to be added.

Finally, heavy and light chain of the antibody are displayed. Each section has its own attributes, displaying the chain’s ID, molecular weight, extinction coefficient, isoelectric point, germline analysis, VGene, DGene, JGene, any clones which use this chain, any plasmids which can be used to construct this chain, and VR Analysis. A sequence map which displays the chain’s sequence along with the annotations associated with the selected pipeline is included. Select different pipelines to view different or more than one set of annotations at a time (i.e. both Kabat and Chothia).

Multispecific antibody detail pages function the same as standard antibody details, with the exception that additional heavy or light chain sections are included depending on the antibody (i.e. a multispecific antibody with two heavy chains and no light chains will only have two heavy chain sequence sections).
